الهدف-  Objectives

TNF-α  was shown to contribute to myocardial contractile dysfunction and cardiomyocyte

death. Since heart diseases are usually associated with alterations in the activity of some ion transporters, this work was  undertaken to investigate the effect of NF-α on the Na+-K+ pump and the Na+K+2Cl- cotransporter of the heart and elucidate its mechanism of action.

 

ألانجازات المحققة   Achievements -

The work demonstrated that TNF-α alters the activity of the Na+-K+ATPase

by inducing PGE2 release through different pathways involving NF-κB and AP-1.

PGE2 then, depending on its concentration, may activate different types of EP receptors and different signaling pathways leading to different effects on the activity of the Na+-K+ATPase

 

آفاق البحث  Perspectives -

With the widespread incidence of congestive heart failure, it becomes imperative to look for still unknown causes and possible treatments.

This   study  uncovers new active molecules that help in  the incidence of heart failure .Understanding the role of TNF-α in heart diseases and  unraveling its signal transduction pathway would help in finding new drugs that could block or exacerbate the effect of the cytokine by interfering  in any of the steps involved in its signaling mechanism. Anti-cytokine therapy studies have already yielded promising results.
